The DRI Flow Cytometry Core Center, directed by Dr. Oliver Umland, provides access to advanced flow cytometry technology to DRI faculty and staff, DRI collaborators, and other investigators within and outside the UM research community. Access may be granted to UM and non-UM researchers whose project align with the DRI mission; to cure diabetes in the fastest and safest way possible.

Dr. Oliver Umland, PhD, Core Director
Dr. Umland earned this Ph.D. in 2003 from the University of Luebeck, Germany. He has more than 15 years of experience in the field of flow cytometry and immunology. He oversees all core functions, including facility policies, consulting investigators regarding experimental and panel design, data analysis and interpretation as well as instrument QC, maintenance, and operation.

Below is a description of the instruments and analyzers at the DRI Flow Cytometry Center:
Beckman Coulter MoFlo Astrios EQ cell sorter
The five laser Beckman Coulter MoFlo Astrios EQ is configured with dual forward scatter PMTs and 355nm (UV), 405nm (violet), 488nm (blue), 561nm (yellow/green), and 638nm (red) excitations lines. The system allows sorting of up to 70000 events per second based on up to 27 physical parameters at the same time with high sensitivity and purity into up to 6 populations. The purchased 100, 200, and 400-micron nozzles enable sorting of very large and/or highly fragile cells including dissociated pancreatic islet cells with extremely high speed. The Cyclone device facilitates precise single cell deposition onto slides and into a wide range of multi well formats suitable for single cell studies, single cell cloning and other downstream applications. This highly flexible instrument allows for small particle sorting and is equipped with a biosafety cabinet to facilitate BSL-2+ sorting.
Beckman Coulter CytoFLEX S
The 15 parameter, 13 color Beckman Coulter CytoFLEX S benchtop analyzer is equipped with 4 lasers (405nm, 488nm, 561nm, and 640nm) and a 96 well plate reader to facilitate automated high throughput analysis. The system provides high sensitivity and resolution with 7 decades of dynamic range and superior linearity and stability. The instrument is capable of resolving 0.22um particles from noise using SSC off the violet laser.
Cytek Aurora
The Cytek Aurora is a full spectral flow cytometer with 5 laser lines (UV, violet, blue, yellow green 561nm, and red). It is equipped with 64 fluorescent and 3 scatter channels and a 96 well plate reader. The unique optical design allows the acquisition of more than 40 colors at the same time and can resolve fluorescent dyes or proteins like for example GFP/YFP that require special filter sets and adjustments on conventional flow cytometers. The instrument is highly sensitive and has a dedicated violet laser side scatter channel for the acquisition of small particles.

The core provides access to 5 Beckman Coulter Kaluza analysis software shared site licenses. These can be used without charge on a first comes first serves basis.
