  Dr. Shiekhattar serves as the Cancer Epigenetics Program Co-Leader at Sylvester Comprehensive Cancer Center. His own laboratory has been interested in understanding the mechanisms by which protein-coding genes are regulated and how disruption of such regulatory mechanisms may lead to neoplasia. Recent evidence has implicated changes in chromatin structure as an essential mechanism in gene regulation. Moreover, besides a classic role for proteins in mediating transcriptional and post-transcriptional effects, it has become clear that non-coding RNAs play an integral role in transcriptional regulation through fine-tuning gene expression patterns during development and differentiation. In the past few years, Dr. Shiekhattar's laboratory has employed a biochemical approach to uncover a number of key factors that mediate transcriptional regulation through modulation of chromatin structure, or post-transcriptional silencing via non-coding RNAs. Further studies are carried out through a detailed structure/function analysis of these factors in regulation of transcription to elucidate the role of these factors in the genesis of cancer.

  Dr. Shiekhattar's lab is rigorously investigating the roles of the RNA polymerase II-associated multiprotein complex, Integrator, in gene expression regulation, epigenetics, and signaling pathways, and how they are manipulated in cancer. They are also exploring the mechanisms of a class of long noncoding RNAs that arise from enhancer elements and behave similar to transcriptional enhancers, in the context of relevant models of development and disease. In the lab, they combine state-of-the-art technologies in genomics, proteomics, and high-resolution imaging.
